To repair a bicycle tire puncture, remove the wheel, use tire levers to get the tire off the rim, remove the inner tube, inflate it to find the hole (by sound or water), scuff the area with the repair kit's buffer, apply vulcanizing fluid (glue), let it dry, press on a patch, and then reinstall the tube and tire, checking for the cause of the puncture in the tire itself before inflating fully.
